Neuropathology in Mice Expressing Human a-Synuclein

چکیده
The presynaptic protein a-synuclein is a prime suspect for contributing to Lewy pathology and clinical aspects of diseases, including Parkinson’s disease, dementia with Lewy bodies, and a Lewy body variant of Alzheimer’s disease. a-Synuclein accumulates in Lewy bodies and Lewy neurites, and two missense mutations (A53T and A30P) in the a-synuclein gene are genetically linked to rare familial forms of Parkinson’s disease. Under control of mouse Thy1 regulatory sequences, expression of A53T mutant human a-synuclein in the nervous system of transgenic mice generated animals with neuronal a-synucleinopathy, features strikingly similar to those observed in human brains with Lewy pathology, neuronal degeneration, and motor defects, despite a lack of transgene expression in dopaminergic neurons of the substantia nigra pars compacta. Neurons in brainstem and motor neurons appeared particularly vulnerable. Motor neuron pathology included axonal damage and denervation of neuromuscular junctions in several muscles examined, suggesting that a-synuclein interfered with a universal mechanism of synapse maintenance. Thy1 transgene expression of wild-type human a-synuclein resulted in similar pathological changes, thus supporting a central role for mutant and wild-type a-synuclein in familial and idiotypic forms of diseases with neuronal a-synucleinopathy and Lewy pathology. These mouse models provide a means to address fundamental aspects of a-synucleinopathy and test therapeutic strategies.

Transgenic mice over-expressing mutant human amyloid precursor protein (PDAPP mouse) develop several Alzheimer’s disease (AD)-like lesions including an age-related accumulation of amyloid-?-containing neuritic plaques. Although aged, heterozygous PDAPP mice also exhibit synaptic and glial cell changes, that is characteristic of AD pathology, no evidence of neurodegeneration has been observed. T...

متن کاملNeuropathology in Mice Expressing Mouse Alpha-Synuclein
α-Synuclein (αSN) in human is tightly linked both neuropathologically and genetically to Parkinson's disease (PD) and related disorders. Disease-causing properties in vivo of the wildtype mouse ortholog (mαSN), which carries a threonine at position 53 like the A53T human mutant version that is genetically linked to PD, were never reported. To this end we generated mouse lines that express mαSN ...

Impaired olfaction is an early pre-motor symptom of Parkinson's disease. The neuropathology underlying olfactory dysfunction in Parkinson's disease is unknown, however α-synuclein accumulation/aggregation and altered neurogenesis might play a role. We characterized olfactory deficits in a transgenic mouse model of Parkinson's disease expressing human wild-type α-synuclein under the control of t...
متن کاملγ-Synucleinopathy: neurodegeneration associated with overexpression of the mouse protein
The role of alpha-synuclein in pathogenesis of familial and idiopathic forms of Parkinson's disease, and other human disorders known as alpha-synucleinopathies, is well established. In contrast, the involvement of two other members of the synuclein family, beta-synuclein and gamma-synuclein, in the development and progression of neurodegeneration is poorly studied. However, there is a growing b...
